:
Alfalfa (Medicago sativa L.) is one of the most important fodder crops in the world and in the Kingdom of Saudi Arabia. Alfalfa is distributed worldwide; adapted and grows in different environments. Investigating the genetic relationships among plant cultivars is of prime importance and assists plant breeders to select cultivars with high yield. This study aims to identifying the biochemical and molecular genetic fingerprints for ten Medicago sativa cultivated in the Kingdom of Saudi Arabia and to elucidate genetic relationships among these cultivars using three types of markers. The first is the random amplified polymorphic DNA (RAPD); the second is the protein markers using polyacrylamide gel electrophoresis (PAGE) and the third is the morphological criteria of the studied cultivars. RAPD-PCR markers generate high levels of polymorphism. The fingerprints generated by twenty arbitrary primers revealed characteristic profiles for each of the ten cultivars. These primers produced a sum of 209 bands, with different sizes ranging from 150 to 2850 base pairs in which 135 bands were polymorphic revealing 64.11% polymorphism, the other 74 bands were monomorphic. Primer OPE-04 generate 100% polymorphism, while Primer OPA-13 produced least percentage of polymorphism which was recorded 20.00%. A total of 41 unique bands were identified out of the polymorphic ones. All the ten cultivars were discriminated by one or more unique bands. Primers OPC-11 and OPE-05 generate largest number of unique bands; each gives 7& 6 unique bands respectively. SDS- polyacrylamide gel electrophoresis technique was used to analyses the storage seed protein of the ten Medicago sativa cultivars. Slight polymorphism (36%) has been identified which could be used as a general fingerprint for these cultivars. A total 25 protein bands were identified in the ten cultivars; 16 bands from them were monomorphic and 9 bands were polymorphic. Morphological data were obtained by measuring 13 morphological characters and recording 56 state characters. The result generated from morphological, RAPD and protein data were analyzed separately and then together using NTSYS-pc software program for drawing the genetic relationships among the Medicago sativa cultivars.
